Abstract

The effect of sperm cryopreservation on DNA fragmentation, viability, and sperm kinetics was evaluated using a Computer Assisted Semen Analysis (C.A.S.A). 76 ejaculates were collected from 5 bulls of the Brown Swiss breed from the National Semen Bank of UNALM. For the analysis of DNA fragmentation, the Sperm-Halomax® Kit was used and for the analysis of the sperm viability Hoeschst 333242 / PI stain. Significant change before and after the cryopreservation for each one of the seminal parameters was evaluated using the T test of related samples, previous verification of normality with the Shapiro-Wilk test. In case data were not normal, the Wilcoxon signed rank test was used. The Bonferroni adjustment was applied for the referred above comparisons. All the analyses were made considering 5 biological replicas in the statistical package SPSS v.23 with 95% reliability. Significant differences were found in 16 of the 21 parameters evaluated, an increase in DNA fragmentation of 3% was evidenced, and decrease in the sperm viability parameter was manifested. Lower values of the following parameters motility, progressive motility, rapid motility was observed, as well as the kinetic parameters VCL, VSL, VAP, DSL, DAP, ALH, BCF, HAC, STR. However higher values of local motility and immobile sperm were observed, attributed to the effect of seminal cryopreservation.
