This work evaluated the plant tissue culture like propagation and preservation alternative of Ilex kunthiana, an endemic shrub. It was found that NaOCl (0,5% by 1 min.) and Benlate®, used as disinfectants of I. Kunthiana seeds, showed the best results in regards to the percentage of seeds adapted to in vitro conditions. Seeds were cultured on the basal medium MS enriched with Gibberellic Acid (GA3) (0,0; 0,5; 1,0 mg/L). When seeds were germinated in vitro, from the seedlings were isolated buds and were cultured in the MS medium enriched with Indol butyric acid (IBA) (0,0; 0,5; 1,0 mg/L) and 6-bencilaminopurina (BAP) (0,0; 0,5; 1,0 mg/L). Subsequently, the regenerated shoots were cultured with Auxins Indol Butyric acid (IBA) (0.0, 0.5, 1,0 mg/L) and Naftalen acetic acid (NAA) (0.0, 0.5; 1,0 mg/L) in order to induce their rooting. It was established that 0.5 mg/l of GA3 facilitated seeds germination (100%) that were the source of buds when were cultured with 0,5 mg/L of BAP and 0,5 mg/L of IBA, obtained the highest number of buds that are then successfully rooted with 0,5 mg/L of IBA establishing better conditions for mass propagation of Ilex kunthiana.
