Culture and characterisation of porcine osteoprogenitor was established in order to study the differentiation of osteoblast in a hybrid material of carbonated apatite (CAp) (patent under process No PI 20042321) and autologous cells that can be used later as an advanced bone graft material. Osteoprogenitors were obtained from enzymatic digestion of fetal porcine periosteum, and were cultured with or without differentiation factors. Biochemical and molecular markers of osteogenesis were obtained. Cells were culture onto CAp of wet and dry synthesis and onto Thermanox (Th) as control substrate. SEM analyses revealed the indistinct adherence and grow of cells cultured on the materials when compared with those cultured on the Th. X-ray diffraction analysis revealed the calcium content of the extra cellular matrix of the cultures. Positive alkaline phosphatase activities and other bone related biochemical and molecular markers were found in time course culture experiments. An animal model for the osteogenesis of periosteal cells was established which demonstrated its use in the evaluation of CAp, indicating that this model is useful to study the functionality of a hybrid material of synthetic CAp and autologous osteoprogenitor cells derived from periosteum.
