TNF-a and IL-10 are regulatory cytokines that participate in the immune response acting either as pro-inflammatory or as anti-inflammatory mediators, respectively. It has been shown that the levels of these mediators influence the susceptibility and the outcome of different infectious, allergic and autoimmune pathologies. An important number of polymorphisms on the promoter region of the corresponding gene have been related with the levels of expression and synthesis of these cytokines, in particular the polymorphism on positions -308 of TNF-a and -1082, -819 and -592 of IL-10. Other studies have demonstrated the association of these single nucleotide polymorphisms (SNP) with the susceptibility and severity of this type of diseases. This review presents the synthesis of the relationship of these genetic polymorphisms with the susceptibility and evolution of infectious, allergic and autoimmune pathologies.
